Subject collection: Metabolic Disorders

  • A MODEL FOR LIFE
Seeking out the sweet spot in cancer therapeutics: an interview with Lewis Cantley
    Lewis C. Cantley
    Disease Models & Mechanisms 2016 9: 911-916; doi: 10.1242/dmm.026856

Summary: We spoke to Lewis Cantley about his career path and the story behind some of his key breakthroughs, including discovery of the phosphoinositide 3-kinase signaling pathway and insights into the role of dysregulated metabolism in cancer.

  • REVIEW
The interplay between obesity and cancer: a fly view
    Susumu Hirabayashi
    Disease Models & Mechanisms 2016 9: 917-926; doi: 10.1242/dmm.025320

Summary: This Review highlights a Drosophila model of diet-induced obesity and cancer, and how these two models are combined to study the interplay between obesity and cancer.

  • RESEARCH ARTICLE
A new glucocerebrosidase-deficient neuronal cell model provides a tool to probe pathophysiology and therapeutics for Gaucher disease
Wendy Westbroek, Matthew Nguyen, Marina Siebert, Taylor Lindstrom, Robert A. Burnett, Elma Aflaki, Olive Jung, Rafael Tamargo, Jorge L. Rodriguez-Gil, Walter Acosta, An Hendrix, Bahafta Behre, Nahid Tayebi, Hideji Fujiwara, Rohini Sidhu, Benoit Renvoise, Edward I. Ginns, Amalia Dutra, Evgenia Pak, Carole Cramer, Daniel S. Ory, William J. Pavan, Ellen Sidransky
    Disease Models & Mechanisms 2016 9: 769-778; doi: 10.1242/dmm.024588

Summary: This work describes the generation of a novel immortalized glucocerebrosidase-deficient neuronal cell model with utility for pathophysiology research and therapeutic development in Gaucher disease.

  • RESEARCH ARTICLE
Increased oxidative stress and apoptosis in the hypothalamus of diabetic male mice in the insulin receptor substrate-2 knockout model
Eva Baquedano, Emma Burgos-Ramos, Sandra Canelles, Agueda González-Rodríguez, Julie A. Chowen, Jesús Argente, Vicente Barrios, Angela M. Valverde, Laura M. Frago
    Disease Models & Mechanisms 2016 9: 573-583; doi: 10.1242/dmm.023515

Summary: Only IRS2-deficient male mice that become diabetic show apoptosis in the hypothalamus and this is associated with an increase in oxidative stress and inflammatory signals. Controlling inflammation and oxidative stress could prevent diabetes-induced brain injury.

  • REVIEW
Using Drosophila to discover mechanisms underlying type 2 diabetes
Ronald W. Alfa, Seung K. Kim
    Disease Models & Mechanisms 2016 9: 365-376; doi: 10.1242/dmm.023887

Drosophila Collection: Studies in Drosophila hold the potential to transform our understanding of the genetics of diabetes. We review important findings and provide a framework for diabetes gene discovery in the fly.

  • RESEARCH ARTICLE
5-HT2A and 5-HT2C receptors as hypothalamic targets of developmental programming in male rats
Malgorzata S. Martin-Gronert, Claire J. Stocker, Edward T. Wargent, Roselle L. Cripps, Alastair S. Garfield, Zorica Jovanovic, Giuseppe D'Agostino, Giles S. H. Yeo, Michael A. Cawthorne, Jonathan R. S. Arch, Lora K. Heisler, Susan E. Ozanne
    Disease Models & Mechanisms 2016 9: 401-412; doi: 10.1242/dmm.023903

Summary: Maternal protein restriction increases fetal brain 5-HT and might contribute to changes in production and function of hypothalamic 5-HT2C and 5-HT2A receptors in the offspring later in life.

  • RESEARCH ARTICLE
A new gestational diabetes mellitus model: hyperglycemia-induced eye malformation via inhibition of Pax6 in the chick embryo
Shi-Jie Zhang, Yi-Fang Li, Rui-Rong Tan, Bun Tsoi, Wen-Shan Huang, Yi-Hua Huang, Xiao-Long Tang, Dan Hu, Nan Yao, Xuesong Yang, Hiroshi Kurihara, Qi Wang, Rong-Rong He
    Disease Models & Mechanisms 2016 9: 177-186; doi: 10.1242/dmm.022012

Summary: Hyperglycemia inhibited Pax6 via oxidative stress and impaired eye development in the chick embryo, a new gestational diabetes mellitus model.

  • RESEARCH ARTICLE
The cannabinoid CB1 receptor and mTORC1 signalling pathways interact to modulate glucose homeostasis in mice
Francisco J. Bermudez-Silva, Silvana Y. Romero-Zerbo, Magalie Haissaguerre, Inmaculada Ruz-Maldonado, Said Lhamyani, Rajaa El Bekay, Antoine Tabarin, Giovanni Marsicano, Daniela Cota
    Disease Models & Mechanisms 2016 9: 51-61; doi: 10.1242/dmm.020750

Summary: Evidence supporting a functional interaction between the endocannabinoid system and the mTORC1 pathway within the endocrine pancreas, which could have implications for the development of new therapeutic approaches for diabetes.

  • RESEARCH ARTICLE
Human umbilical cord matrix-derived stem cells exert trophic effects on β-cell survival in diabetic rats and isolated islets
Yunting Zhou, Qi Hu, Fuyi Chen, Juan Zhang, Jincheng Guo, Hongwu Wang, Jiang Gu, Lian Ma, Guyu Ho
    Disease Models & Mechanisms 2015 8: 1625-1633; doi: 10.1242/dmm.021857

Summary: The newly identified paracrine role of umbilical cord stem cells highlights their therapeutic utilities to treat not only the autoimmune-mediated type 1 diabetes but also type 2 diabetes.
